Research Funding

I have consistently submitted applications to the NIH and the March of Dimes to fund my research program. I have successfully secured R01 funding from the NIH during a time when there has been a significant decrease in the number of grants funded as well as substantial changes to the grant submission process. I was also successful in receiving a competitive administrative supplement to expand the experimental approaches proposed within the parent R01 application. Since I began writing grants in 2006, there has been a reduction in the number of revised applications from two to one. Subsequent submissions require significant changes to the scope. During this time there was also a reduction in the length of the proposal from 25 pages to 12. The pay lines at the National Institute of Allergy and Infectious Diseases, where the majority of my UPEC grants have been assigned) has consistently been reduced by at least one percentile point each year that I have submitted applications from 14% in 2006 to 8% in 2013. Success in achieving funding has required substantial adaptation, creativity and innovation to remain competitive. To this end, I have continued to strengthen my independent research programs as well as have establish strong and productive collaborations to compliment my expertise and provide balanced and comprehensive approaches to unveil mechanisms associated with bacterial virulence and disease. I have also worked diligently to improve my grantsmanship skills through formal courses, discussions with other faculty. In addition, serving as an ad hoc reviewer has provided valuable insight and additional examples of successful and unsuccessful grants.



I have completed a contract from AstraTech to conduct a clinical trail based upon our experimental evidence to determine the effects of catheter type on the microbiota present in the urinary tract of patients with lower spinal cord dysfunction. This trial was performed in conjunction with the Division of Urology. We have also obtained intramural funding to conduct an FDA approved Phase I trial to evaluate the efficacy of a novel non-antibiotic, non-vaccine approach for the prevention of urinary tract infections.



I have been successful in obtaining both intramural and extramural funding for my trainees. Drs. Doug Storm and Ashay Patel, my first two Pediatric Urology Fellows, obtained intramural funding to perform the research in my laboratory. Dr. Elizabeth Lucas obtained extramural funding from the Thrasher Research Foundation to support her research program in my laboratory. She received the award with her first submission. Kristin Greathouse received an F31 to support her stipend during her PhD training in Nursing to be performed in my laboratory. Unfortunately, Kristin elected to perform more clinically based research in another laboratory for her dissertation after receipt of the award.



Although I have received NIH funding, I have not decreased my effort to fund the additional projects in my laboratory. Two R01 applications were submitted to the NIH in June and will be reviewed in October. I will submit another NIH grant in the fall cycle to further our clinical trials to evaluate prognostic and preventative technologies to combat urinary tract infections. It was always been, and continues to be, my intent to maintain my research program with at least two nationally reviewed granting mechanisms.

Biographical

My passion for microbiology began during my training to become a Dental Hygienist. As an undergraduate, I participated in the conduct of two clinical trials, which provided me with the unique opportunity, at an early stage in my development, to observe the translation of intellectual concepts into improved patient care. During the years that followed my graduation in 1986 with Bachelors in Science with Distinction from the University of Missouri-Kansas City, I felt gratified with helping patients improve their oral health. However, I soon realized that I wanted to do more through a mechanistic understanding of infectious disease. My experiences during my undergraduate training as well as working in private practice provided the scaffold that would shape my scientific career.



I continued my education at the University of Missouri-Kansas City with the completion of my Masters of Science in Biochemistry in 1994. Under the guidance of Dr. Joan Stader, my Masters thesis focused on the assembly of an inner membrane protein complex that is required for transport of nascent proteins produced in the cytoplasm to the periplasm or outer membrane of Escherichia coli. My graduate education concluded at the University of Connecticut Health Sciences Center. In the laboratory of Dr. Lawrence Rothfield, I investigated protein complexes that regulate cell morphology and division in E. coli. I attained my PhD in Biochemistry and Molecular Biology in 2000. My combined graduate studies provided me with a strong foundation in bacterial genetics, protein biochemistry, and microscopy.



I returned to my interest in bacterial pathogenesis through my postdoctoral training in the laboratory of Dr. Scott Hultgren at Washington University in Saint Louis. I was awarded a Ruth L. Kirschstein National Research Service Award Individual Postdoctoral Fellowship to investigate the role of bacterial morphologic differentiation and intracellular community development as essential components of bacterial pathogenic lifestyles using the disease model of uropathogenic E. coli-mediated urinary tract infections.



I became an Assistant Professor of Pediatrics in 2006 and the within the Division of Urology in 2010. With regard to mechanisms of disease, I have become an internationally recognized expert in the role of bacterial morphology and the architectural attributes of bacterial community development, particularly of intracellular populations during infection. I also have expertise in the role of early proinflammatory responses of the epithelium and the influence of these effectors on bacterial physiology and disease progression. With regard to evidence-based approaches to improve patient management, I have identified bacterial traits that are prognostic of disease severity of urinary tract infection and disease sequelae as well as a novel non-antibiotic, non-vaccine approach for the prevention of urinary tract infections.
