Advising Narrative

I have mentored 18 undergraduate students, 12 of whom published papers with us in high impact journals. Several of my students were accepted in Medical Schools, PhD programs, nursing schools, physical therapy, dentistry and speech pathology programs. They succeeded in pursuing their career aims with the help of the training that they have gained in my lab and for having the skills to contribute as co-authors on prominent publications. In addition many of them gained awards and fellowships. Youssra Saqr obtained the Cystic Fibrosis summer fellowship while Seeshidra Hoque received the Ohio State University STEP award. In addition, I have trained 2 students from the SUCCESS program in OSU which allows students from disadvantaged and underrepresented groups to receive scientific training during the summer. I consider training undergraduate students the most important step in the foundation of an enthusiastic researcher interested in studies in medicine.

Postdoctoral / Researcher Advising

As my laboratory expanded and my funds increased, postdoctoral fellows started contacting me to join my laboratory. My first postdoc was Dr. Sheetal Kotrange. Her project was focused on the inflammatory response in macrophages. She published a first author publication and then left to join clinical training. Then, Dr. Benjamin Kopp joined my lab as a fellow from Nationwide Children's Hospital (NCH). His work in my lab was focused on Burkholderia-mediated defective killing mechanisms in macrophages from CF patients. During his training, he learned how to perform and interpret scientific experiments. He obtained a K08 training award under my mentorship. He is now a well-funded physician scientist at NCH. Recently, Kyle Caution joined my lab as a postdoctoral fellow after obtaining his PhD with my group. His novel project as a postdoc is focused on cystic fibrosis. He obtained a Training Grant from the Cystic Fibrosis Foundation and a T32 Training grant which he had to decline. He has been invited to give a talk at the Immunity meeting in Australia. He published 12 co-author papers, 1 first author manuscript in Nature Scientific Reports, 2 reviews and 2 book chapters. I was also contacted by Kathrin Krause who expressed her desire to join my lab for her postdoctoral training after obtaining her PhD from Germany. She is supported by a 2 year fellowship from the German Government. She has been in my lab for 16 months and she has published a review, a book chapter and is a co-author on our paper in Nature Scientific reports. She is now working on her first author publication. It is exciting and thrilling to observe how these postdoctoral fellows develop into well rounded independent scientists.

Published Works

With more than 15 years experience in the field of innate immunity, host-pathogen interaction and autophagy, I have been intensely engaged in exciting and novel studies to understand how the innate immune system recognizes and responds to pathogens and how mutations in these sensing systems lead to infectious and inflammatory diseases in humans. I have made important contributions to the field of Nod like receptors, inflammasomes and autophagy field and ongoing studies are investigating the sophisticated system governing the activation of signaling cascades and the mechanisms involved. My accomplishments include 53 publications including papers in high impact journals such as Plos Pathogens (IF 9.0) 2009, Immunity (IF 24) 2012, Autophagy (IF 12.5) 2012, 2016, Journal of Biological Chemistry (IF 4.5) 2011, 2013, Frontiers in Microbiology (IF 5.5) and Nature Scientific Reports 2016 (IF 5.5). My report in Plos Pathogens discussed a novel role for caspase-7 in regulation of the intracellular trafficking of bacteria-containing vacuoles. This paper is the first to show a non-apoptotic function for caspase-7. My papers in Journal of Biological Chemistry and Frontiers in Microbiology demonstrated how an adaptor molecule called ASC modulates intracellular bacterial infection in macrophages. I have a strong reputation in the field of Legionella pneumophila pathogenesis, a bacterium that causes pneumonia in the elderly and immune compromised. My recent papers in European Journal of Immunology and Frontiers in Cellular and infection Microbiology improved our understanding of how Legionella derived from biofilms infect human immune cells. They also demonstrate how this intracellular pathogen interacts with the autophagy pathway. These reports led to the discovery that autophagy activity is compromised in old mice and has been published in Epigenetics Journal offering a previously unrecognized mechanism for weak autophagy in the aging population. My most recent publication in Nature Scientific Reports provides mechanistic insight into how caspase-1 and caspase-11 perform opposing functions to modulate the actin machinery and trafficking in macrophages, therefore controlling intracellular infection.

As one of the most active groups in the field autophagy, host pathogen interaction and inflammasome, we have obtained and developed a large collection of tools and animal models to investigate signaling pathways affecting autophagy, caspases, the actin machinery, innate immunity and inflammation. Therefore, our current studies logically build on our prior work on autophagy, innate immunity and inflammation. Yet, it is innovative since the underlying factors leading to weak autophagy in several disease conditions and extensive inflammation in others are unclear. Recently, I have characterized the innate immune deficiency in macrophages derived from cystic fibrosis patients. My publications were subjects for press release. I published papers in Autophagy, JBC, Journal of Leukocyte Biology demonstrating an unprecedented role for autophagy in the pathobiology of cystic fibrosis. These papers were the first to show that cystic fibrosis is a disease with an underlying immune deficiency. This was followed by our recent publication in Autophagy Journal discerning the mechanism of autophagy reduction in cystic fibrosis macrophages. We reported the existence of a cluster of microRNAs that is highly expressed in CF cells and that normalization of the expression levels of this cluster, corrects autophagy and the function of the CFTR chloride channel.

In addition, my trainees have submitted more than 70 abstracts to several local, national and international meetings.

Therefore, my research is focused on areas that are relevant to human diseases accompanying infection, immunity and inflammation and my work is well perceived, highly cited and well recognized.

My h-index is 26, i10-index 37 and I have more than 8,000 citations. "Please note that The Research in View system does not update citations".

Master/Graduate Advising

I started my laboratory at Ohio State University with great motivation to perform high quality science that will impact human disease conditions and train excellent scientists. I started with one laboratory technician and a graduate student. In a short time, 2 funded graduate students joined my group. I consider the graduation of my first student Dalia Abdelaziz a gratifying achievement. She was awarded several travel awards and awards for best presentations with multiple publications. She published several important papers on the inflammasome. Then, my second student Basant Abdelrahman gained more recognition when she was invited to give a talk at an International meeting besides the travel awards and awards for excellent talks. She was first author on seminal publications in the CF field. As I gained more experience in encouraging graduate students to seek awards and scientific recognition, my student Mia Tazi accomplished her PhD in less than 5 years and obtained 15 awards ranging from travel awards to a 2 year fellowship from the American Association of Immunologists. She has 9 papers besides her first author publication in Autophagy Journal (IF 12.5). In parallel, my other graduate student Kyle Caution flourished as a superb scientist with more than 12 papers including a first author paper in Nature Scientific Reports, a review and 2 book chapters. He has obtained 11 awards including a 2 year fellowship from the Cystic Fibrosis Foundation. I attribute their outstanding performance to 2 major reasons. First, this is due to the excellent caliber of graduate students admitted to PhD biological and biomedical programs at OSU. Second, it is due to the dedicated well rounded collaborative training that I offer in my laboratory. I train them to be independent thinkers besides skilled technicians. I thrive to teach them how to present their ideas in a simple and comprehensive way with several practices and attending meetings to present their work. I instigate their perseverance and desire to succeed. I also boost their self-confidence and independence. My approach has been rewarding and successful as demonstrated by the numerous awards they have obtained along the years. The collaborative common space setting of our department tremendously aided in the success of my students.
